Oil. The whole story was an analogy for oil and the geopolitics of the time. Spice allowed shipping to occur, you could become "addicted" to it, etc. The desert world being the only place you could find it, and the Houses of the Landsraad subduing the local population to extract it, basically occupying the planet.
